Permselectivity of the glomerular wall examined with iron compound tracer.

چکیده
Rat kidney endothelial cell morphology was examined after introducing iron colloid particles of positive or negative charge to investigate the relationship between the electric charge and permeation through the glomerular capillary. The kidneys were first perfused with Hanks’ solution through the renal arteries and then with iron colloid particles of positive or negative charge. The iron colloid particles of positive charge were prepared with ferric chloride and cacodylate solutions, and the negative particles were prepared with iron chondroitin sulfate colloid particles. The iron colloid particles of positive charge adhered to the surface of endothelial cells of the glomerular capillaries, as well as the arterioles, capillaries and venules. Some particles were taken up by pinocytosis, accumulated in the glomerular basement membrane and appeared in the urinary spaces passing through the filtration slits of podocytes. Iron colloid particles of negative charge neither adhered to the endothelial cells nor were taken by the cells. They did not permeate into the urinary spaces. Permeation into the tubular lumen through the peritubular venules was not observed with particles of positive or negative charge. ∗PMID: 143189 [PubMed indexed for MEDLINE] Copyright c ©OKAYAMA UNIVERSITY MEDICAL SCHOOL Acta Med. Glomerular handling of native albumin in the presence of circulating modified albumins by the normal rat kidney.
Persistent hyperglycemia, as occurring in diabetes, induces changes in circulating as well as in structural proteins. These changes involve substitution of lysine residues by glucose adducts resulting in early Amadori products that evolve into toxic and active substances, the advanced glycation end adducts. In previous studies, we demonstrated that early glycated (Amadori) albumin infused into ...

To examine the electrostatic effects of fixed negative charges on the glomerular capillary wall, polydisperse [(3)H]DEAE dextran, a polycationic form of dextran, was infused into 10 Munich-Wistar rats. Fractional clearances of DEAE ranging in radius from 18 to 44A were determined in these rats, together with direct measurements of the forces and flows governing the glomerular filtration rate of...

To explore the mechanism of glomerular permselectivity defect in passive Heymann nephritis, an experimental model of human membranous glomerulopathy, Munich-Wistar rats were subjected to both micropuncture assessment of glomerular hemodynamics and whole kidney clearance measurements of graded size dextrans 10 days after injection of sheep anti-rat tubular antigen (anti-Fx1A). Compared with norm...

Conclusion It would appear then, that both GBM and the renal parenchyma as a whole contain diverse charged components. Abnormalities of GBM anionic charge expression are more complex than at first conceived, reflecting often subtle alterations in membrane matrix charged structures which may have a minimal or profound, direct or indirect influence on membrane permeability. The original concept o...
متن کاملPermselectivity of the glomerular capillary wall to macromolecules. I. Theoretical considerations.
The transport of macromolecules across the renal glomerular capillary wall has been described theoretically using flux equations based on (a) restricted transport through small pores, and (b) the Kedem-Katchalsky formulation. The various assumptions and limitations inherent in these two approaches are discussed. To examine the coupling between macromolecular solute transport and the determinant...
